Lean vs. Obese Mice: The Ventral Prostate Revisited International Journal of Morphology
Martins,Fabiane Ferreira; del Sol,Mariano; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto.
Obese mice (C57BL/6J-ob/ob) do not express leptin and develops hyperphagia, decreased energy expenditure, obesity, hyperglycemia, hyperinsulinemia, hypothermia, and infertility. Obesity causes reproductive dysfunction with negative impacts on prostatic structure and fertility. We aimed to compare the structure and molecular aspects of the ventral prostate between of lean and obese (ob/ob) mice. Three months old male lean and obese mice had their prostates dissected and prepared for light microscopy and immunofluorescence. In comparison to the lean mouse, the obese mouse showed a substantial structural modification in the ventral prostate starting with an atrophy of the prostate ventral lobe. Histologically, the acini showed a reduction in size, and in the...

Marine Polysaccharide-Collagen Coatings on Ti6Al4V Alloy Formed by Self-Assembly ArchiMer
Norris, Karl; Mishukova, Oksana; Zykwinska, Agata; Colliec-jouault, Sylvia; Sinquin, Corinne; Koptioug, Andrei; Cuenot, Stéphane; Kerns, Jemma; Surmeneva, Maria; Surmenev, Roman; Douglas, Timothy.
Polysaccharides of marine origin are gaining interest as biomaterial components. Bacteria derived from deep-sea hydrothermal vents can produce sulfated exopolysaccharides (EPS), which can influence cell behavior. The use of such polysaccharides as components of organic, collagen fibril-based coatings on biomaterial surfaces remains unexplored. In this study, collagen fibril coatings enriched with HE800 and GY785 EPS derivatives were deposited on titanium alloy (Ti6Al4V) scaffolds produced by rapid prototyping and subjected to physicochemical and cell biological characterization. Coatings were formed by a self-assembly process whereby polysaccharides were added to acidic collagen molecule solution, followed by neutralization to induced self-assembly of...

Methylation-specific PCR analysis in Col8A1 promoter in Creole cattle carrier of rob(1;29) Electron. J. Biotechnol.
Postiglioni,Alicia; García,Cristina B; Rincón,Gonzalo; Arruga,M. Victoria.
Robertsonian translocation (rob(1;29)) is the most frequent structural chromosomal abnormality in cattle. Heterozygous carriers have a normal phenotype but show a 3-5% decrease in fertility. Chromatin decondensation was evaluated similar to the inactive X chromosome when submitted to demethylating agent. Based on this result, and the concept that imprinted genes are essential in embryonic development, we decided to query genes located on BTA1 and BTA29 that could undergo genome imprinting. The collagen typeVIII-α1 (Col8A1) acted on extracellular matrix structural proteins. DNA bisulfite conversion and sequentiation methods were used to compare its differential methylation patterns. It was performed on eight Creole cattle DNA blood samples from normal and...

Modulation of extracellular matrix by nutritional hepatotrophic factors in thioacetamide-induced liver cirrhosis in the rat BJMBR
Guerra,R.R.; Trotta,M.R.; Parra,O.M.; Avanzo,J.L.; Bateman,A.; Aloia,T.P.A.; Dagli,M.L.Z.; Hernandez-Blazquez,F.J..
Nutritional substances associated to some hormones enhance liver regeneration when injected intraperitoneally, being denominated hepatotrophic factors (HF). Here we verified if a solution of HF (glucose, vitamins, salts, amino acids, glucagon, insulin, and triiodothyronine) can revert liver cirrhosis and how some extracellular matrices are affected. Cirrhosis was induced for 14 weeks in 45 female Wistar rats (200 mg) by intraperitoneal injections of thioacetamide (200 mg/kg). Twenty-five rats received intraperitoneal HF twice a day for 10 days (40 mL·kg-1·day-1) and 20 rats received physiological saline. Fifteen rats were used as control. The HF applied to cirrhotic rats significantly: a) reduced the relative mRNA expression of the genes: Col-α1 (-53%),...

Osteogenic molecules for clinical applications: improving the BMP-collagen system Biol. Res.
Arrabal,Pilar M; Visser,Rick; Santos-Ruiz,Leonor; Becerra,José; Cifuentes,Manuel.
Among the osteogenic growth factors used for bone tissue engineering, bone morphogenetic proteins (BMPs) are the most extensively studied for use in orthopaedic surgery. BMP-2 and BMP-7 have been widely investigated for developing therapeutic strategies and are the only two approved for use in several clinical applications. Due to the chemical and biological characteristics of these molecules, their authorised uses are always in combination with a carrier based on collagen type I. Although the use of these growth factors is considered safe in the short term, the very high doses needed to obtain significant osteoinduction make these treatments expensive and their long-term safety uncertain, since they are highly pleiotropic and have the capacity to induce...

Placental osseous metaplasia associated with abortion in a mare: case report Arq. Bras. Med. Vet. Zootec.
Lima,P.A.; Eckstein,C.; Ecco,R.; Santos,R.L..
ABSTRACT The aim of this report is to describe a case of marked osseous metaplasia in the placenta associated with abortion in a mare. A 19-year-old Campolina mare with a history of multiple parturitions and absence of reproductive failure, aborted a fetus with approximately six months of gestation. There were no gross changes in the fetus and the placenta. Microscopically, there was multifocal replacement of fetal connective mesenchyme of the chorionic villi with well differentiated and partially mineralized osseous tissue (confirmed by von Kossa staining). To further characterize the collagenous component of the lesion, sections of the placenta were stained with Masson’s trichrome and Picrosirius red. Osseous tissue deposition had abundance of type I...

Porcine Uterine and Placental Extracellular Matrix Molecules Throughout Pregnancy International Journal of Morphology
Sanchis,E. G; Cristofolini,A. L; Taglialegna,A; Merkis,C. I.
The molecules that constitute the extracellular matrix are important in several functions related to tissue support and cell-cell, cell-extracellular matrix interaction. Among the macromolecules that constitute the mentioned matrix we find osteopontin, fibrinogen and collagen. The present study was undertaken to analyze the rol of osteopontin, fibrinogen and collagen in uterine-placental interface during normal porcine gestation. Uterine and placental tissues from crossbred gilts of 30 (n=5), 60 (n=5), 70 (n=5) and 114 (at term, n=5) days of gestation were used. Macroscopic analysis of the embryos/fetuses allowed us to determine their gestational age by means of the crown-rump lenght. Haematoxylin-Eosin and Masson's Trichrome dyes along with light...

Quantitative Analysis of Collagen Fibrillar Structure in Canine Uterus Exposed to Acetate of Medroxyprogesterone Using Second Harmonic Generation Microscopy International Journal of Morphology
Salinas,Paulo; Miglino,Maria Angélica; del Sol,Mariano.
Collagen is the most abundant structural protein found in organs and is responsible for providing tissues with structure and function. In order to investigate in canine uteri the potential effect of medroxyprogesterone acetate (MPA) on the changes in collagen deposition were grouped as nulliparous (n=11), multiparous (n=11) and treated with MPA (n=11; nulliparous with two treatments; 5 mg/kg; i.m.). The amount of collagen was studied in the fold and basal regions of the endometrium and myometrium using second harmonic generation with a two-photon spectral confocal microscope, quantified using ImageJ software with a color segmentation plugin, was expressed as fraction area (%) and analyzed by ANOVA (p<0,05). No differences were observed between groups in...

Review of the Y chromosome and hypertension BJMBR
Ely,D.; Turner,M.; Milsted,A..
The Y chromosome from spontaneously hypertensive rats (SHR) has a locus that raises blood pressure 20-25 mmHg. Associated with the SHR Y chromosome effect is a 4-week earlier pubertal rise of testosterone and dependence upon the androgen receptor for the full blood pressure effect. Several indices of enhanced sympathetic nervous system (SNS) activity are also associated with the SHR Y chromosome. Blockade of SNS outflow reduced the blood pressure effect. Salt sensitivity was increased by the Y chromosome as was salt appetite which was SNS dependent. A strong correlation (r = 0.57, P<0.001) was demonstrable between plasma testosterone and angiotensin II. Coronary collagen increased with blood pressure and the presence of the SHR Y chromosome. A...

Soft Collagen-Gelatine Sponges by Convection Drying BABT
Meyer,Michael; Trommer,Kristin.
This study showed that thermally labile fibrillar collagen could be processed continuously in combination with gelatine as foaming additive by convection drying. The procedure led to stable sponges with similar structural and physical properties as found for freeze-dried collagen samples. The fibrillar collagen remained native, while gelatine acted as foaming additive. The absorbency of the sponges was improved by opening the surface with abrasives. A use as medical device with hemostyptic properties would be possible.

Supraorganized collagen enhances Schwann cell reactivity and organization in vitro BJMBR
Maturana,L.G.; Zanon,R.G.; Pierucci,A.; Vidal,B.C.; Oliveira,A.L.R..
We investigated the reactivity and expression of basal lamina collagen by Schwann cells (SCs) cultivated on a supraorganized bovine-derived collagen substrate. SC cultures were obtained from sciatic nerves of neonatal Sprague-Dawley rats and seeded on 24-well culture plates containing collagen substrate. The homogeneity of the cultures was evaluated with an SC marker antibody (anti-S-100). After 1 week, the cultures were fixed and processed for immunocytochemistry by using antibodies against type IV collagen, S-100 and p75NTR (pan neurotrophin receptor) and for scanning electron microscopy (SEM). Positive labeling with antibodies to the cited molecules was observed, indicating that the collagen substrate stimulates SC alignment and adhesion (collagen IV...

The extracellular matrix of the lung and its role in edema formation Anais da ABC (AABC)
Pelosi,Paolo; Rocco,Patricia R.M.; Negrini,Daniela; Passi,Alberto.
The extracellular matrix is composed of a three-dimensional fiber mesh filled with different macromolecules such as: collagen (mainly type I and III), elastin, glycosaminoglycans, and proteoglycans. In the lung, the extracellular matrix has several functions which provide: 1) mechanical tensile and compressive strength and elasticity, 2) low mechanical tissue compliance contributing to the maintenance of normal interstitial fluid dynamics, 3) low resistive pathway for an effective gas exchange, d) control of cell behavior by the binding of growth factors, chemokines, cytokines and the interaction with cell-surface receptors, and e) tissue repair and remodeling. Fragmentation and disorganization of extracellular matrix components comprises the protective...

The in vivo effect of L-arginine on skin elasticity in mice BJPS
Souza,Ádria do Prado Barros de; Oliveira,Marina Miranda Rocha de; Andrade,Rafael Rocha de; Amorim,Rivadávio Fernandes Batista de; Bocca,Anamélia Lonrezetti; Borin,Maria de Fátima.
ABSTRACT The human skin aging process is a complex mechanism that can be induced both by intrinsic and extrinsic factors. Observations include a decrease in the biosynthetic and proliferative capacity of cells, increased expression of matrix metalloproteinases, reduction in collagen type I expression, and the progressive disappearance of elastic tissue in the papillary dermis. L-arginine, the substrate of nitric oxide synthesis, is involved in angiogenesis and cell proliferation, as well as an indirect precursor of collagen synthesis via the proline pathway. The aim of this study was to examine the tensile strength, histology, and immunohistochemistry of female and male mice skin receiving different concentrations of topically applied L-arginine, in order...

ULTRASTRUCTURAL ANALYSIS OF PINEALECTOMY AND LACK OF LIGHT INFLUENCE OVER COLLAGEN IN THE ENDOMETRIUM OF RATS International Journal of Morphology
Pinto de Medeiros,Juliana; Wanderley Teixeira,Valéria; Aguiar Coelho Teixeira,Álvaro; Baratella Evêncio,Liriane; Evêncio Neto,Joaquim.
Some studies have shown that pinealectomized female rats present a large concentration of collagen fibers in the endometrium. The lack of light results in morphologic alterations in the uterine lamina propria and in the muscular layer, reducing thus the uterine weight. Therefore, this work aimed to analyze ultra-structurally the influence of melatonin in the remodeling of the collagen in the endometrium, in the estrus phase, in pinealectomized adult rats or submitted to lack of light. The experimental groups: I- sham-pinealectomized maintained in a clear/dark cycle 12/12 hours, during three months; II- pinealectomized rats maintained in a clear/dark cycle 12/12 hours, for three months; III- normal rats maintained in clear/dark cycle 12/12 hours for three...

Unusual Glycosaminoglycans from a Deep Sea Hydrothermal Bacterium Improve Fibrillar Collagen Structuring and Fibroblast Activities in Engineered Connective Tissues ArchiMer
Senni, Karim; Gueniche, Farida; Changotade, Sylvie; Septier, Dominique; Sinquin, Corinne; Ratiskol, Jacqueline; Lutomski, Didier; Godeau, Gaston; Guezennec, Jean; Colliec-jouault, Sylvia.
Biopolymers produced by marine organisms can offer useful tools for regenerative medicine. Particularly, HE800 exopolysaccharide (HE800 EPS) secreted by a deep-sea hydrothermal bacterium displays an interesting glycosaminoglycan-like feature resembling hyaluronan. Previous studies demonstrated its effectiveness to enhance in vivo bone regeneration and to support osteoblastic cell metabolism in culture. Thus, in order to assess the usefulness of this high-molecular weight polymer in tissue engineering and tissue repair, in vitro reconstructed connective tissues containing HE800 EPS were performed. We showed that this polysaccharide promotes both collagen structuring and extracellular matrix settle by dermal fibroblasts. Furthermore, from the native HE800...
